To understand the mechanism of transcriptional regulation of Periplaneta fuliginosa densovirus(PfDNV)nonstructural genes,5 -flanking sequence of ns3 gene(325 bp),which encompasses the region from the 5 -terminus of the viral genome to the translation initiation codon of the ns3 gene,was cloned.
为了研究蟑螂Periplanetafuliginosa浓核病毒(pfDNV)非结构基因转录调控的机制,用PCR的方法从蟑螂浓核病毒基因组DNA中扩增了非结构蛋白基因ns3翻译起始密码子上游325bp的启动子序列;并进一步以其作为模板,利用聚合酶链式反应技术,得到6个不同长度的启动子片段和两个定点突变体片段,构建由其驱动的萤火虫荧光素酶基因报告质粒,在脂质体介导下转染多种昆虫细胞,体外分析了该启动子的活性。

妊娠期人细小病毒感染
人类细小病毒可引起某些感染，但大多较轻微。孕妇感染该病毒后一旦传给胎儿，因胎儿免疫功能尚不完善，常可导致严重感染；人细小病毒可抑制胎儿红细胞成熟系统，从而引起胎儿贫血、缺氧、充血性心力衰竭，最终导致胎儿流产、死产或积水型死胎。因此，近年来国外对孕妇中人细小病毒的感染颇为关注，并已建立了相应的检测技术，如EIA、RIA、ELISA、免疫荧光、核酸杂交和PCR等。
