从广义上讲植物抗病基因(resistance gene)与防御反应基因(defense gene)都是在植物抗病反应过程中起抵抗病菌侵染及扩展的有关基因。所谓抗病基因就是flor经典遗传学基因对基因(gene-for-gene)假说中所指的与病原菌无毒基因相对应的,存在于植物特定品种中,在植物生长的整个周期或其中某个阶段为组成型表达的植物抗病品种所特有的一类基因。植物防御反应基因的特点是,在抗病和感病品种中均存在,其差异主要体现在基因表达的时间、空间及产物含量的不同,为组成型或诱导型表达的一类基因。
植物抗病基因工程所首选的最佳目的基因是来自植物自身的抗病基因。但由于植物的基因组十分庞大,加之人们对抗病基因产物的功能了解甚少,以及克隆手段的限制,直到1992年以后,才出现成功克隆抗病基因的报道,使植物抗病基因工程出现了质的飞跃。
