[方法]以从乐山峨边花牛骨骼肌中提取总RNA为模板,利用RT-PCR技术扩增获得肌肉生长抑制素基因(MSTN)cDNA片段,将其克隆到T载体上,通过PCR鉴定阳性克隆并对其进行测序鉴定。

本研究以我国主要地方山羊品种及部分引入品种为研究对象,利用PCR技术,扩增了山羊肌肉生长抑制素基因(GenBank登录号为DQ167575),测序后对其序列的结构、核苷酸组成及编码的氨基酸进行了初步研究,并利用Bioedit、Phylip等生物学软件构建了山羊MSTN基因进化树,同时对各品种山羊MSTN基因部分序列进行了PCR-RFLP分析。
