最敏感的受试动物接触某种毒物，用已知最敏感的观察指标或用现代的检查方法测出该毒物能引起受试动物产生异常生理、生化等反应或潜在的病理学改变的最小剂量。毒物的这一最小剂量称为阈剂量；如毒物的最小剂量是以浓度表示的，则称为阈浓度。阈剂量（或阈浓度）以下的剂量为阈下剂量（或阈下浓度）。在阈下剂量的作用下，用现代检查方法不能观察到机体的任何异常生理、生化反应或潜在的病理学改变。在实际应用中只选择阈下剂量中一个最大的剂量，这一剂量称为最大无作用剂量（或最大无作用浓度）。一般所说的阈下剂量就是指最大无作用剂量。
从阈剂量开始到刚好引起受试动物死亡的剂量为止（实际工作中以半数致死量为上限），为毒物的毒作用带。毒物的毒作用带范围越小，危险性就越大。
阈剂量是确立最大无作用剂量的依据；最大无作用剂量又是确立有害物质在环境中的最大容许浓度的毒理学依据。
