Then we will design and construct the eukaryotic expression vector of human Par-4 gene, transfect it into human leukemia cells K562, investigate the effect of Par-4 to WT1mRNA and protein expression level in K562, and finally observe the influence of Par-4 gene up regulation on K562 biological behaviour such as cell proliferation, apoptosis and cell cycle.
本实验拟采用荧光定量RT-PCR及Western blotting的方法,首先观察Par-4基因、WT1基因在急性白血病及非白血病患者骨髓细胞中的表达情况,并进一步探讨其在白血病细胞凋亡过程中的表达变化。

Methods Based on PC12 cell apoptosis induced by H_2O_2, cell survival rate was determined by MTr colorimetric analysis,prostate apoptosis response-4(Par-4)mRNA ex- pression and nuclear factor-kappa B(NF-κB)mRNA expression determined by reverse transcription-polymerase chain reaction(RT-PCR),Par-4 and NF-κB protein expression determined by Western blot method.
方法在H_2O_2诱导PC12细胞凋亡模型的基础上,采用MTT比色分析检测细胞存活率,RT-PCR检测前列腺凋亡响应基因Par-4(prostate apoptosis response-4,Par-4)和核转录因子-κB p65(nuclear factor-kappaB,NF-KB)mRNA表达,Westem blot检测Par-4和NF-κBp65蛋白表达。

提取新生鼠脑组织总RNA,利用RT-PCR的方法扩增Brn-4的基因片段,用基因重组技术将Brn-4基因片段克隆到真核表达载体pEGFP-C2中,测序及PCR进行鉴定;将重组质粒应用脂质体转染的方法转入MSC中,荧光显微镜观察绿色荧光蛋白(GFP)的表达。
